Abstract Дисертаційна робота спрямована на вивчення морфофункціональних особливостей кісткового мозку статевозрілих щурів і хімічного складу гемопоетичної тканини за умов дії на організм важких металів та обґрунтування можливості корекції їх пошкоджувальної дії вітаміном Е. Для комплексного вивчення варіабельності гемопоетичної тканини та крові в умовах мікроелементозу було визначено дизайн дослідження, що включав використання 84 статевозрілих білих щурів-самців, яких було поділено на групи та серії згідно з умовами й тривалістю спостереження. Під час гістологічного, гістохімічного та імуногістохімічного досліджень препаратів кісткового мозку щурів виявлено, що надмірне споживання солей важких металів призводить до зменшення площі еритроцито- та гранулоцитопоезу за одночасного зростання кількості мегакаріоцитів. Одночасно відбуваються виражені коливання якісних показників гемопоетичної тканини, що виявляються дистрофічно-некротичними та початковими диспластичними змінами в усіх його складових. Гематотоксична дія проявляється порушенням функціональної здатності кісткового мозку у вигляді коливання показників загального аналізу крові, які безпосередньо залежать від терміну споживання важких металів та ступеня їх накопичення. За рахунок додавання вітаміну Е в терапевтичних дозах до раціону щурів спостерігалося зменшення негативних морфологічних змін у кістковому мозку та крові впродовж уживання солей важких металів, а також прискорення регенераторних можливостей кровотворної тканини в період відновлення.
Диссертационная работа направлена на изучение морфофункциональных особенностей костного мозга половозрелых крыс и химического состава гемопоэтической ткани при действии на организм тяжелых металлов и обоснование возможности коррекции их повреждающего действия витамином Е. Для комплексного изучения вариабельности гемопоэтической ткани и крови в условиях микроэлементоза был определен дизайн исследования, включающий использование 84 беспородных половозрелых белых крыс-самцов, которые были распределены на группы и серии согласно условиям и продолжительности наблюдения. При гистологическом, гистохимическом и иммуногистохимическом исследованиях препаратов костного мозга крыс выявлено, что чрезмерное потребление солей тяжелых металлов приводит к уменьшению площади эритроцито- и гранулоцитопоэза при одновременном росте количества мегакариоцитов. Одновременно происходят существенные колебания качественных показателей гемопоэтической ткани, проявляющиеся дистрофическинекротическими и начальными диспластическими изменениями во всех его составляющих. Гематотоксическое действие проявляется нарушением функциональной способности костного мозга в виде колебания показателей общего анализа крови, непосредственно зависящих от срока потребления тяжелых металлов и степени их накопления. За счет добавления витамина Е в терапевтических дозах в рацион крыс наблюдались уменьшение негативных морфологических изменений в костном мозге и крови на протяжении употребления солей тяжелых металлов, а также ускорение регенераторных возможностей кроветворной ткани в период восстановления.
For a complex study of the effects of heavy metals, a design of research was identified which included the use of 84 pubescent white male rats. In order to determine the action of pollutants on the bone marrow and the restorative capacity of hematopoietic tissue, animals had been into two groups divided. Addition of vitamin E to the animal diet was the basis for dividing all the animals into series in order to study the protective properties of the corrector. The study of the main results of the experiment and the rationale for using the corrector for preventive-therapeutic purposes were performed at the expense of modern research methods (microscopic examination of morphological changes of bone marrow in histological, histochemical and immunohistochemical slides, their morphometry, quantitative evaluation of indicators of the chemical composition of hematopoietic tissue, study of indicators of general and biochemical analysis of blood, statistical processing of digital data). It was found that the addition of heavy metals to the diet of animals leads to their accumulation in hematopoietic tissue. This is accompanied by a decrease in the area of the erythrocyto- and granulocytopoiesis while increasing the number of megakaryocytes. Moreover, there are significant fluctuations in the qualitative composition of the bone marrow, which are manifested by dystrophic-necrotic and initial dysplastic changes in all its components. We found that the haematotoxic action is manifested by a violation of the functional capacity of the hematopoietic tissue in the form of fluctuations in indicators of general blood analysis, which are directly dependent on the period of consumption of heavy metals and their degree of accumulation. Due to the use of a complex of modern methods, the features of bone marrow readaptation capabilities have been established, provided that xenobiotics are discontinued. The expressed restorative possibilities of hematopoietic tissue, which also depend on the term of observation, are shown. It was found that even within 90 days after the abolition of the pollutants, there was no complete removal of heavy metals from the body that turned out a deviation from the norm of qualitative and quantitative indicators of myeloid tissue and blood. For the first time, under the experimental conditions of excessive intake of heavy metals salts, the protective properties of vitamin E were studied for restoration of normal structure and function of bone marrow of experimental animals. By adding it at therapeutic doses to the diet of rats, there was a decrease in negative morphological changes in the bone marrow and blood during the use of heavy metal salts, as well as accelerating the regenerative capacity of hematopoietic tissue during the recovery period. The complex analysis of the obtained results made it possible to clarify and supplement information on the pathogenetic links of the negative influence of heavy metals on the process of hematopoiesis, which is realized through the direct and indirect influence of exogenous pollutants on various sprouts of hematopoiesis. A clear link between the hematotoxic effect of xenobiotics and the functioning of the hematopoietic blood system was revealed. On the basis of the results of a comprehensive experimental study, the structural features of the bone marrow of rats were clarified and its key differences from human hematopoietic tissue were determined, both in normal and during the consumption of xenobiotics. This may be the basis for further modeling of changes in hematopoietic tissue when studying the effects of damaging factors. The revealed results demonstrate in detail the fluctuations of hematopoiesis and blood indices during consumption of a mixture of heavy metal salts. The established changes clearly demonstrate the pronounced hematotoxic effect of these xenobiotics upon their excessive admission to the body.
