    The number of patients with diabetes increases annually. Modern forecasts predict that diabetes will be the seventh leading cause of death in 2030. Despite many significant advances in the research of diabetes and the use of new modern treatments, the disease is still progressing, and it is necessary to continue to study the effects of diabetes on human systems and organs: kidney and myocardium.

    Diabetes mellitus is a global medical and social problem. 2-3% of the population suffers from diabetes mellitus. According to current forecasts, the number of people with diabetes mellitus will double every 15 years. Diabetes mellitus is the basis for the development of complex comorbidities and complications such as diabetic microangiopathy and neuropathy. Diabetes patients are at increased risk for atherosclerosis, coronary heart disease, and renal failure. Scientists around the world have been thoroughly engaged in the problem of hyperglycemia, which is the consequence of type 1 diabetes mellitus, but the causes of its occurrence have not been fully established yet. The study aimed to investigate the lungs of pre-reproductive age rats under the influence of alloxan-induced hyperglycemia.

    Exploring the advances of single-cell RNA sequencing in thyroid cancer: a narrative review

    Thyroid cancer, a prevalent form of endocrine malignancy, has witnessed a substantial increase in occurrence in recent decades. To gain a comprehensive understanding of thyroid cancer at the single-cell level, this narrative review evaluates the applications of single-cell RNA sequencing (scRNA-seq) in thyroid cancer research. ScRNA-seq has revolutionised the identifcation and characterisation of distinct cell subpopulations, cell-to-cell communications, and receptor interactions, revealing unprecedented heterogeneity and shedding light on novel biomarkers for therapeutic discovery. These fndings aid in the construction of predictive models on disease prognosis and therapeutic efcacy. Altogether, scRNA-seq has deepened our understanding of the tumour microenvironment immunologic insights, informing future studies in the development of efective personalised treatment for patients. Challenges and limitations of scRNA-seq, such as technical biases, fnancial barriers, and ethical concerns, are discussed. Advancements in computational methods, the advent of artifcial intelligence (AI), machine learning (ML), and deep learning (DL), and the importance of single-cell data sharing and collaborative eforts are highlighted. Future directions of scRNA-seq in thyroid cancer research include investigating intra-tumoral heterogeneity, integrating with other omics technologies, exploring the non-coding RNA landscape, and studying rare subtypes. Overall, scRNA-seq has transformed thyroid cancer research and holds immense potential for advancing personalised therapies and improving patient outcomes. Eforts to make this technology more accessible and cost-efective will be crucial to ensuring its widespread utilisation in healthcare.

    Histoultrastructural features of thymocytes due to t he impact of the experimental general dehydration of a mild degree

    ABSTRACT The aim: The objective of the current study was to reveal ultrastructural changes in rats’ thymocytes in experimental data in conditions of mild general dehydration. Materials and methods: The study was conducted on 20 non-linear adult male laboratory rats weighing 150-170 g. Histological and semi-thin slides of the thymus were prepared according to the required guidelines. Results: On average, in the cortical zone of the thymus, there was decreased cellularity by 13.4% (p<0.001), while in the medulla zone this indicator turned out to be unreliable - 5.5% (р=0.19), compared to the indicators in animals of the control group. The study showed that a slight degree of general dehydration of the body causes ultrastructural changes in the thymus and is accompanied by a cell-mediated response of the central link of immunogenesis and results in morphological changes in the thymus, which are atrophic in nature with a typical pattern of remodeling of the organ’s microstructure, which corresponds to cellular aging and the associated sign of accelerated involution. Conclusions: General dehydration of a mild degree in the experiment is accompanied by a cell-mediated response of the central link of immunogenesis and results in morphological changes in the thymus, which are atrophic in nature with a typical pattern of remodeling of the organ’s microstructure, which corresponds to cellular aging and the associated sign of accelerated involution.

Scientists around the world have been thoroughly engaged in the problem of hyperglycemia, which is the consequence of type 1 diabetes mellitus, but the causes of its occurrence have not been fully established yet. The study aimed to investigate the lungs of pre-reproductive age rats under the influence of alloxan-induced hyperglycemia. Material and Methods: 12 rats of 3 months of age were involved in the experiment. They were divided into two series: experimental and control. The Control series includes six intact rats. For the experimental modeling of hyperglycemia, we used alloxan monohydrate, pre-dissolved in 0.9% solution of sodium chloride, which was then injected intraperitoneally once at a dose of 20 mg/100 g on an empty stomach. The experiment lasted 60 days. We measured the glucose level using the Accu-Chek Advantage (Boehringer, Germany) at2, 12, and 24 hours after alloxan injection, and then weekly. The average level of glucose in the blood remained at 11 mmol/L ± 2 mmol/L. To study the remodeling of the lungs, we used the histological examination by a light microscope (Olympus BH-2) and histomorphometry using the universal certified program “SEO Scan Lab 2.0” and “SEO Image Lab 2.0”. The obtained data were processed on a personal computer using the software “GraphPad” and a licensed Excel XP spreadsheet editor (Microsoft Office 2013). The data were analyzed by unpaired t-test. P values ≤0.05 were considered as statistically significant. Results: Obtained morphometric data shows the increase of the width of alveoli by 8.5% (p < 0.05), the width of the conducting part of the respiratory bronchiole by 15.7 % (p < 0.05) and the thickness of the interalveolar septum by 8.3% (p < 0.05). The wall thickness of pulmonary vessels increased by 22.9% (p <0.05). Lymphoid nodules were more hypertrophied compared to the control ones by 2.4 times (p <0.05). Conclusions: Alloxan-induced hyperglycemia leads to the development of emphysematous changes in combination with restriction. The vascular wall of muscular pulmonary vessels was thickened due to hypertrophy of the tunica media and tunica externa. The hypertrophy of lymphoid follicles and lymph nodes of the upper and lower mediastinum was observed in experimental animals.

    Prolonged hypoosmolar overhydration causes a volume overload necessitating cardioprotectors together with a complex of measures aimed at stabilizing the water and electrolyte balance. The content of water and electrolytes in the human body changes throughout life, so the body's response to the disorder of water and salt balance may be different depending on age. The fluid overload positively correlates with adverse outcomes in critically ill patients. One of the main tasks of modern cardiology is to find the means of leveling the influence of adverse factors on myocardium in different aged patients. The aim of our study was the determination of morphological changes in the myocardium of old laboratory animals under the influence of severe hypoosmolar hyperhydration and attempt to correct the identified changes by means of meldonium.

    Histoultramicroscopic Investigation of the Rats’ Thymus (Experimental Data)

    The research paper presents the characteristic of cytoarchitectonics of the thymus of intact white mature male laboratory rats. Topicality of the study is due to the need to clarify the data on the contribution of each type of thymus cells in the formation of its structure. The aim of the research was to determine the specifics of localization and ultramicroscopic structure of thymus cells in male mature Wistar laboratory rats. Material and Methods: The study was conducted using histological and ultramicroscopic methods on 10 mature male laboratory rats, weighing 130-150 g. Semi-thin (0.5-1 μm) and ultrathin (0.05-0.2 μm) sections were made on a microtome UMTP-4 (Ukraine), which were stained with 1% methylene blue solution with the addition of 1% sodium tetraborate solution. Histological analysis and photographic recording were performed using Olympus light microscope (Japan) and DSM 510 camcorder with magnification in 1000 times. Results: With a detailed study of the semi-thin and ultrathin sections in the thymus lobules the specifics of localization and ultramicroscopic structure of thymus cells were clearly identified. The features of localization and ultramicroscopic structure of epithelial, mesenchymal, vascular and hematopoietic thymus cells were determined from the point of view of their functional loads and interactions. Conclusion: The described structural peculiarities of the components of the thymus and their relative location in different zones reflect significant organ polymorphism, which must be taken into account in order to achieve the required level of objectivity in the result evaluation of simulated biomedical experiments.

    Оrganometric changes in thymus under the influence of propylene glycol

    Наукова зацікавленість впливу ксенобіотиків на організм людини викликана тим, що імунним органам властива яскраво виражена відповідна реакція на дію ендогенних і екзогенних факторів. Останнім часом вивчення імунологічних порушень, як проявів реакцій на екопатогенні умови, відводиться основна патогенетична роль в розвитку серцево-судинних, нервово-психічних захворювань і дифузних захворювань сполучної тканини. Мета дослідження - визначення органометрічних змін в тимусі самців щурів під впливом пропіленгліколя. 40 статевозрілих щурів самців лінії WAG були розділені на дві групи: I контрольну групу склали 8 тварин, II основну групу - 32 щури, розділені на 4 підгрупи по 8 гризунів в кожній, яким через шлунковий зонд вводили водний розчин пропіленгліколю в дозі 1/10 ДЛ50 в перерахунку на 26,38 г / кг протягом 7, 15, 30, 45 днів. Після декапитации тварин відповідно до терміну, встановленого в експерименті, витягувався тимус і вимірювалися лінійні розміри (довжина, ширина, висота) за допомогою цифрового штангенциркуля, а також визначалась маса і обсяг органу. Розраховані межі мінливості морфометричних показників тимуса в інтактній та експериментальних групах. На підставі проведеного дослідження встановлено, що вплив пропиленгликоля викликає органометричні зміни в тимусі щурів. Однак більш виражені зміни спостерігалися на 7 і 30 добу. Встановлено такі межі коливань індексів індивідуальної мінливості: IndHLтімуса в експериментальній групі коливалася в межах від min = 12,57 до max = 47,54, середнє значення склало від 24,67 до 28,02; IndHWтімуса був в межах від min = 11,96 до max = 88,73, середнє значення - 36,78-41,41; IndT експериментальної групи - в межах від min = 38,17 до max = 141,3, середнє значення складало 71,1-86,52. Вивчення морфометричних параметрів тимуса в експериментальній групі щурів дозволило встановити достовірні зниження всіх параметрів і їх відхилення від показників контрольної групи, що свідчить про активну реакцію тимуса на індукований ксенобіотик.

    Age-specific features of cardioprotective action of meldonium under the experimental severe hypoosmolar hyperhydration

    Важкий ступінь гіпоосмолярної гіпергідратації призводить до значного збільшення масометричних показників серця молодих і старих експериментальних щурів. Застосування мельдонію уповільнює збільшення маси правого шлуночка і його дилатацію в усіх вікових категоріях.
