    Morphological analysis of papillary thyroid carcinoma with psammoma bodies

    Background & Objective: During last decade, the proportion of thyroid cancer, among other types of cancer had a clear tendency to rise. Papillary thyroid carcinoma (PTC) is the most common cancer of this organ, constituting up to ca. 70% cases. For the majority of patients the general survival rate depends on the histological features of the tumour and on many other predictors, one of which is pathological biomineralization. The aim of the work is to carry out morphological analysis of the tissues papillary thyroid carcinoma with psammoma bodies. Method: We have analysed the samples from 54 PTC patients and controls by using immunohistochemistry and spectroscopic techniques. The samples were divided into two groups: the PTC group included 24 cases of PTC with psammoma bodies and the control group was constituted of 30 PTC cases without psammoma bodies and other manifestations of pathological biomineralization. Results: We have demonstrated the clear colocalization of osteopontin and calprotectin in the psammoma bodies and suggested the model for their laminated structure development. Immunostaining with of activated Caspase 3 antibodies revealed significantly higher number of apoptotic cells in the samples of PTC with PBs. Conclusion: We have found intensive immunostaining with osteopontin antibodies in the tumour tissues and in the tumour surrounding, which indicates that osteopontin may counteract biomineralization. We have shown that the major component of PBs is hydroxyapatite.

    Osteonectin overexpression in the case of prostate cancer with intraluminal inclusions

    Osteonectin (OSN) is secreted by osteoblasts during bone formation, initiating mineralization and promoting mineral crystal formation at sites of ectopic calcification. Also OSN was found in many types of human malignant tumours. The aim is to study the OSN expression in patients with prostate cancer (PC) and the presence of intraluminal inclusions (prostatolithes and amyloid cells). Method: OSN expression was investigated in tumours and in the adjacent prostatic tissue of 30 PCs with intraluminal inclusions and 30 PCs without them by immunohistochemistry. In each group 15 samples refer to moderately differentiated G2 and low-differentiated G3 tumours. Samples were fixed, embedded in paraffin, and analized for OSN accumulation using the anti-OSN antibody, followed by DAB detection substrate and counterstained with Mayer’s haematoxylin. Results: OSN expression was increased in PC tissues with pathological inclusions in comparison to those without them (p<0.001, Student test). Osteonectin was mostly localized in tumour cells cytoplasm, its expression was not observed in tumour microangiourea cells and in stroma. It was found that the OSN expression by tumour cells reduced during reduction of the malignant tumour differentiation (comparison of subgroups G2 and G3) (p<0.001 and p<0.01 respectively for groups I and II). Conclusion: OSN overexpression in tumours and in the adjacent prostatic tissue of PCs with intraluminal inclusions may be regarded as a prospective role for the osteosteogenic phenotype development of tumour cells and for the bone metastasis promotion.

    T-lymphocytes and periodontal inflammation (short report)

    Об'єктом даного дослідження є аналіз експресіїCD3 позитивних клітин в пародонті при запаленні. Методи.27 пацієнтів з гігантоклітинним епулісом і30 пацієнтів з гострим і хронічним запаленням оцінювали на наявністьCD3 позитивних клітин імуногістохімічно. Результати. Гігантські клітини не єCD3 позитивними, тоді як тільки23,28% клітин інфільтрату в гігантоклітинному епулісі були СD3 позитивними(P <0,05). При гострому запаленні клітинний інфі-льтрат мав32,7 ± 2,43% СD3 позитивних клітин. CD3 иммунореактивность була56,2 ± 8,6% у клітин-ному інфільтраті(р<0,05) при хронічному запаленні. Висновок. Хронічні запалення пародонту характеризується наявністю в сполучній тканині рясної Т-клітинної запальної інфільтрації.

    Identification of epulis patients with a high risk of periodontal diseases: DNA infrared spectra, expression MGMT and p53 by and apoptosis analysis

    Background The objective of this study was to epulis apoptosis analyze, expression of MGMT and p53, DNA methylation patients with a high risk of developing bone resorption and inflammatory periodontal diseases. Methods. Human epulis benign were assessed for their ability to expression of MGMT, p53. 51 epulis DNA were compared by infrared spectroscopy for their ability to apoptosis induced. We conducted a comparative analysis of global expression changes in human epulis using K-S test and Student method . Results. MGMT and p53 was expressed in giant-cell. Expression of MGMT and p53 in giant-cell epulis. By immunohistochemistry, 97.1±0.42% (P<0.001) of giant-cells were positive for MGMT, whereas only 6.21±0.26% of giant-cells were positive for p53 (P<0.001). Induction of the enzymatic activity of MGMT was increased p53. This could be explained by the fact that MGMT is physiologically expressed by the giant-cell of epulis, p53 expression is weak or absent in giant-cell epulis and non induced apoptosis in the fibrous connective tissue. Significant changes were observed IR absorption bands at - δsSN3 group bases. In intact periodontium in CH3 - IR bands were unchanged. Center band oscillations - δsSN3 group was at 1375 ± 1cm - 1. Percentage intensity on a scale transmission of infrared radiation was 7,18 ± 0,74%. Percentage of infrared absorption bands 1375 ± 1 cm- 1 in giant cell epulis equal to 13,24±3,7% (** P = 0.01). Changes in IR absorption bands observed in δsCH2 group. In intact periodontal δsCH2 next strip - strip center fluctuations - 1464 cm- 1, the percentage of intensity on a scale transmission of infrared radiation - 0,24 ± 0,03%. Percentage of IR absorption bands - 1464 cm-1 in the giant cell epulis was 0,46±0,09% (* P = 0.05). Percent transmittance intensity on a scale stretching, deformation and rocking vibrations of CH3 and CH2 groups of DNA Conclusion. Aim of this study was to find if IR can be used in neoplasm process DNA methylation detection. Statistical analysis shows that there are differences between spectra of giant cell epulis and control group. Fitting analysis allows to follow small changes in spectra CH3 groups. Presented results prove that infrared spectroscopy could be useful tool in DNA methylation. Morphologically, apoptosis is characterized by DNA fragmentation and formation of apoptotic. DNA CH3 groups can be detected by infrared spectra leads of breaks in DNA strands. This leads to the activation of apoptosis via p53 and MGMT.

    Periodontitis and atherosclerosis – mechanisms of association through matrix metalloproteinase 1 expression

    Wprowadzenie. Od dawna jest postulowany związek między zapaleniem przyzębia a miażdżycą naczyń i jej powikłaniami. Cel pracy. Analiza ekspresji metaloproteinazy 1 (MMP-1) w nabłonku dziąsłowym i podnabłonkowej tkance łącznej u osób, które zmarły z powodu powikłań miażdżycy naczyń. Materiał i metody. Zbadano 20 wycinków dziąsła osób, które zmarły z powodu powikłań miażdżycy w szpitalu w S umie na Ukrainie. Grupę kontrolną stanowiło 9 wycinków dziąsła osób zmarłych z innych przyczyn. Ekspresję MMP-1 oceniano za pomocą badania immunohistochemicznego i badania wykonanego z zastosowaniem mikroskopii fluorescencyjnej. Wyniki. Immunoekspresja MMP-1 w grupie z miażdżycą była potwierdzona przez brązowe zabarwienie cytoplazmy keratynocytów w nabłonku dziąsłowym i komórkach nacieku leukocytarnego w podnabłonkowej tkance łącznej. Ekspresja MMP-1 w warstwach nabłonka dziąsłowego dotyczyła 95,8 ± 2,43% keratynocytów i była istotnie
