Modern forecasts predict that diabetes will be the seventh leading cause of death in 2030. Despite many significant advances in the research of diabetes and the use of new modern treatments, the disease is still progressing, and it is necessary to continue to study the effects of diabetes on human systems and organs: kidney and myocardium.Item Histoultrastructural features of thymocytes due to t he impact of the experimental general dehydration of a mild degree(Aluna Publishing, 2023) Приходько, Ольга Олексіївна; Prykhodko, Olha Oleksiivna; Avilova, O.; Дмитрук, Сергій Миколайович; Dmytruk, Serhii Mykolaiovych; Понирко, Аліна Олексіївна; Ponyrko, Alina Oleksiivna; Гордієнко, Олена Володимирівна; Hordiienko, Olena Volodymyrivna; Prykhodko, E.ABSTRACT The aim: The objective of the current study was to reveal ultrastructural changes in rats’ thymocytes in experimental data in conditions of mild general dehydration. Materials and methods: The study was conducted on 20 non-linear adult male laboratory rats weighing 150-170 g. Histological and semi-thin slides of the thymus were prepared according to the required guidelines. Results: On average, in the cortical zone of the thymus, there was decreased cellularity by 13.4% (p<0.001), while in the medulla zone this indicator turned out to be unreliable - 5.5% (р=0.19), compared to the indicators in animals of the control group. The study showed that a slight degree of general dehydration of the body causes ultrastructural changes in the thymus and is accompanied by a cell-mediated response of the central link of immunogenesis and results in morphological changes in the thymus, which are atrophic in nature with a typical pattern of remodeling of the organ’s microstructure, which corresponds to cellular aging and the associated sign of accelerated involution. Conclusions: General dehydration of a mild degree in the experiment is accompanied by a cell-mediated response of the central link of immunogenesis and results in morphological changes in the thymus, which are atrophic in nature with a typical pattern of remodeling of the organ’s microstructure, which corresponds to cellular aging and the associated sign of accelerated involution.Item Correction of myocardial changes on an animal experimental model with meldonim(Oxford University Press, 2022) Ярмоленкo, Ольга Сергіївна; Ярмоленко, Ольга Сергеевна; Yarmolenkо, Olha Serhiivna; Бумейстер, Валентина Іванівна; Бумейстер, Валентина Ивановна; Bumeister, Valentyna Ivanivna; Приходько, Ольга Олексіївна; Prykhodko, Olha Oleksiivna; Гордієнко, Олена Володимирівна; Гордиенко, Елена Владимировна; Hordiienko, Olena Volodymyrivna; Деміхова, Надія Володимирівна; Демихова, Надежда Владимировна; Demikhova, Nadiia VolodymyrivnaProlonged hypoosmolar overhydration causes a volume overload necessitating cardioprotectors together with a complex of measures aimed at stabilizing the water and electrolyte balance. The content of water and electrolytes in the human body changes throughout life, so the body's response to the disorder of water and salt balance may be different depending on age. The fluid overload positively correlates with adverse outcomes in critically ill patients. One of the main tasks of modern cardiology is to find the means of leveling the influence of adverse factors on myocardium in different aged patients. The aim of our study was the determination of morphological changes in the myocardium of old laboratory animals under the influence of severe hypoosmolar hyperhydration and attempt to correct the identified changes by means of meldonium.Item Influence of antitumor chemotherapeutics on mechanical properties of injured tubular skeletal bone(Азіз Алієв, Азербайджанський державний інститут удосконалення лікарів, 2022) Рябенко, Тетяна Василівна; Рябенко, Татьяна Васильевна; Riabenko, Tetiana Vasylivna; Дмитрук, Сергій Миколайович; Дмитрук, Сергей Николаевич; Dmytruk, Serhii Mykolaiovych; Ярмоленкo, Ольга Сергіївна; Ярмоленко, Ольга Сергеевна; Yarmolenkо, Olha Serhiivna; Пернаков, Микола Станіславович; Пернаков, Николай Станиславович; Pernakov, Mykola Stanislavovych; Теслик, Тетяна Петрівна; Теслик, Татьяна Петровна; Teslyk, Tetiana Petrivna; Понирко, Аліна Олексіївна; Понырко, Алина Алексеевна; Ponyrko, Alina Oleksiivna; Гордієнко, Олена Володимирівна; Гордиенко, Елена Владимировна; Hordiienko, Olena VolodymyrivnaЦель работы – изучить влияние противоопухолевых химиопрепаратов на механические свойства травмированной длинной кости скелета. Item Effect of L-tocopherol on morphological reformations of rat pineal gland under the impact of heavy metal salts(Poltava state medical university and Ukrainian public organization Scientific society of anatomists, histologists, embryologists and topographoanatomists of Ukraine, Kiptenko, Liudmyla IvanivnaThe article presents the results of the study on morphological rearrangements of structural changes in the rat pineal gland under the influence of heavy metal salts and correction with L-tocopherol during 30 days. General morphological and statistical research methods were used (histological, morphometric and statistic methods). The protective effect of L-tocopherol caused in the pineal gland a number of compensatory-adaptive processes aimed at neutralizing the stressor’s impact on the organ. The 30- days protective period of L-tocopherol is insufficient to completely neutralize the impact of heavy metals on the organ: the glial response was activated, vascular area increased, vascular wall permeability was impaired, edema was formed and the connective tissue component growth was taking place, indicating dysfunctional reorganization in the gland’s secretory activity. The above morphological changes negatively affected the processes of hormones evacuation into the blood, the course of the general adaptation syndrome and the homeostasis restoration in the organ.Item The effect of the experimental chronic hyperglycemia on the kidney and myocardium(State Institution «Institute of Nephrology NAMS of Ukraine", 2021) Ярмоленкo, Ольга Сергіївна; Ярмоленко, Ольга Сергеевна; Yarmolenkо, Olha Serhiivna; Бумейстер, Валентина Іванівна; Бумейстер, Валентина Ивановна; Bumeister, Valentyna Ivanivna; Polak, S.; Гордієнко, Олена Володимирівна; Гордиенко, Елена Владимировна; Hordiienko, Olena Volodymyrivna; Приходько, Ольга Олексіївна; Prykhodko, Olha Oleksiivna; Деміхова, Надія Володимирівна; Демихова, Надежда Владимировна; Demikhova, Nadiia Volodymyrivna; Шкатула, Юрій Васильович; Шкатула, Юрий Васильевич; Shkatula, Yurii Vasylovych; Деміхов, Олексій Ігорович; Демихов, Алексей Игоревич; Demikhov, Oleksii Ihorovychhe number of patients with diabetes increases annually. Modern forecasts predict that diabetes will be the seventh leading cause of death in 2030. Despite many significant advances in the research of diabetes and the use of new modern treatments, the disease is still progressing, and it is necessary to continue to study the effects of diabetes on human systems and organs: kidney and myocardium.Methods. A total of 24 rats of reproductive age (6 months old) were involved in this experimental study. Experimental rats were injected with alloxan intraperitoneally once at a dose of 20 mg/100 g on an empty stomach. In addition, they received a 10% glucose solution 24 hours after alloxan injection and a 5% glucose solution during the experiment. We measured glucose level with Accu-Chek Advantage (Boehringer, Germany) after 2, 12, and 24 hours after alloxan injection, and then weekly. The subjects of the investigation were kidney and heart of the experimental (n=12) and control (n=12) animals for correct comparative analysis.Results. The average blood glucose level remained at 11 mmol/L ± 2 mmol/L. During the experimental period, the rats’ weight gain, dilation of both ventricles and relative renal weight gain were determined. By the histological examination of the myocardium, we revealed polymorphic nuclei, perinuclear cytolysis, fragmentation, wavy-like deformation of cardiomyocytes, stromal and perivascular edema, uneven filling of blood vessels, and local fibrosis. Thinning of fibrous capsule and cortical layer, destruction of nephrons, and hemorrhages were detected in the kidney. Conclusions. Our study confirms the robustness of alloxan-induced hyperglycemia in rats. We came to this conclusion because the early changes in the kidneys and heart are explained by the development of microangiopathies, which is a typical feature of the pathogenesis of diabetes. With prolonged exposure to chronic hyperglycemia, structural disorders of vital organs are worsened. This experimental model could be used for conducting comprehensive research aimed to study the mechanisms of diabetes mellitus, the effects of hyperglycemia on organs and tissues, and correct the complications. X-ray diffraction analysis helps to determine age-related features of hydroxyapatite structure in a spongy bone tissue. Comparing the obtained data with literature data (6,7,8), the ontogenetic transformations in spongy bone tissue can be divided into several periods: 1) period of intensive osteogenesis - in immature age; 2) stabilization period is in mature animals; 3) period of regressive changes with the predominance of resorption processes in old rats. The mineral component of the calcaneus matrix in laboratory rats of different age groups is characterized by changes at the microstructural level. An increasing in crystallite size rises the level of microdeformation of the crystal lattice. Defective increase, i.e. structural imperfection of biominerals, indicates violations of normal metabolic processes in the calcaneus, which in turn leads to a decrease in its strength.Item Histoultramicroscopic Investigation of the Rats’ Thymus (Experimental Data)(Duzce University Medical School, 2021) Приходько, Ольга Олексіївна; Prykhodko, Olha Oleksiivna; Дмитрук, Сергій Миколайович; Дмитрук, Сергей Николаевич; Dmytruk, Serhii Mykolaiovych; Avilova, O.; Kubikova, E.; Бумейстер, Валентина Іванівна; Бумейстер, Валентина Ивановна; Bumeister, Valentyna Ivanivna; Ярмоленкo, Ольга Сергіївна; Ярмоленко, Ольга Сергеевна; Yarmolenkо, Olha Serhiivna; Гордієнко, Олена Володимирівна; Гордиенко, Елена Владимировна; Hordiienko, Olena VolodymyrivnaThe research paper presents the characteristic of cytoarchitectonics of the thymus of intact white mature male laboratory rats. Topicality of the study is due to the need to clarify the data on the contribution of each type of thymus cells in the formation of its structure. The aim of the research was to determine the specifics of localization and ultramicroscopic structure of thymus cells in male mature Wistar laboratory rats. Material and Methods: The study was conducted using histological and ultramicroscopic methods on 10 mature male laboratory rats, weighing 130-150 g. Semi-thin (0.5-1 μm) and ultrathin (0.05-0.2 μm) sections were made on a microtome UMTP-4 (Ukraine), which were stained with 1% methylene blue solution with the addition of 1% sodium tetraborate solution. Histological analysis and photographic recording were performed using Olympus light microscope (Japan) and DSM 510 camcorder with magnification in 1000 times. Results: With a detailed study of the semi-thin and ultrathin sections in the thymus lobules the specifics of localization and ultramicroscopic structure of thymus cells were clearly identified. The features of localization and ultramicroscopic structure of epithelial, mesenchymal, vascular and hematopoietic thymus cells were determined from the point of view of their functional loads and interactions. Conclusion: The described structural peculiarities of the components of the thymus and their relative location in different zones reflect significant organ polymorphism, which must be taken into account in order to achieve the required level of objectivity in the result evaluation of simulated biomedical experiments.
